摘要:The microscale distribution of fluorescently labeled cellulose enzyme provides quantitative information on the location of amenable and recalcitrant sites on particles of pretreated woody biomass. Statistical colocalization of the labeled enzyme with cellulose and lignin provides a quantitative measurement of enzyme interaction with woody cell walls showing a surprisingly diverse range of interactions among individual biomass particles. Cellulase enzyme was more strongly correlated to cellulose distribution in cellulose-rich secondary walls compared to lignin-rich middle lamella regions. The distribution of cellulose accounts for 30% of the variation in enzyme distribution after correction for the correlation between cellulose and lignin distribution. Comparison of steam exploded pine with pine holocellulose suggests that enzyme binding is strongly associated with particle surfaces with little evidence for infiltration by enzyme even in highly porous biomass particles. This study contributes to understanding the role of pretreatment in enzymatic hydrolysis of recalcitrant softwood biomass for sustainable biofuel applications.
